Simvastatin and Alendronate Effects against Cholangiocarcinoma Cells

New treatments are needed for cholangiocarcinoma, an aggressive cancer with a poor prognosis. Combination of statins and bisphosphonates may be useful in treating through influencing the mevalonate pathway. We studied synergistic effects simvastatin alendronate against cholangiocarcinoma cells, looking first at cytotoxic, apoptotic anti-migratory effects, then possible mechanisms action. Individually, both increased cell death combination was more effective. Additionally, treatment inhibited colony formation, reduced protein-related proliferation, NF-κB cyclin D1, p53 death. Apoptosis induced by activation early cells stimulation caspase 3 enzyme. The also decreased migration suppressing matrix metalloproteinases, metalloproteinase-2 metalloproteinase-9 levels. Finally, Rac1 protein expression Our study shows that plus has direct anticancer effect on vitro. Mevalonate inhibitors could used as therapeutic options perhaps, chemotherapy-sensitizing agents.
